Solifenacin succinate (Vesicare) is a once-daily oral tablet used to treat overactive bladder (OAB) with symptoms of urge urinary incontinence, urgency, and urinary frequency. As a selective muscarinic receptor antagonist, it reduces involuntary bladder muscle contractions, decreasing the frequency of urgent and incontinent episodes. The usual starting dose is 5 mg once daily, which may be increased to 10 mg once daily based on individual response and tolerability; the dose should not exceed 5 mg daily in patients with severe renal or hepatic impairment or in those taking strong CYP3A4 inhibitors. Common side effects include dry mouth, constipation, blurred vision, and dry eyes. Solifenacin should be used with caution in patients with decreased gastrointestinal motility, urinary retention, controlled narrow-angle glaucoma, or QT prolongation risk factors, and is contraindicated in uncontrolled narrow-angle glaucoma, urinary retention, or gastric retention. Rare but serious angioedema of the face, lips, tongue, and/or larynx has been reported and requires immediate discontinuation and medical attention.
